One of the Mayflower passengers, Standish played a leading role in the administration and defense of Plymouth Colony from its start. On February 17, 1621, he was officially voted the first commander of the Plymouth Colony Militia and was continually re-elected to that position for the remainder of his life. Later, he served as an agent of Plymouth Colony in England, as assistant governor, and as treasurer of Plymouth Colony. He was also one of the first settlers and founders of the town of Duxbury, Massachusetts.

Dès le débarquement du Mayflower, il saffaira à lorganisation de la défense de la colonie. Le 17 février 1621, il était élu commandant de la colonie. Par la suite, il fut député de Plymouth auprès de la Couronne, et exerça les fonctions de gouverneur suppléant et de trésorier de la colonie. Il fut l'un des fondateurs de la ville de Duxbury (du nom de la terre ancestrale de Standish à Duxbury Woods, dans le district de Chorley ) en 1632.
